Course Objective This laboratory course is concerned with molecular and biochemical aspects of enzymology involving purification and structural/functional characterization of enzymes.
Course Content This laboratory course is concerned with molecular and biochemical aspects of enzymology which is the purification and structural/functional characterization of enzymes. Techniques are homogenization, to obtain cell free extract, centrifugation to separate membrane from soluble proteins, initial separations by ammonium sulfate precipitation and purifications by chromatography (anion/cation exchange, hydrophobic interaction, gel filtration, HPLC, and several concentration methods). Assessment protocols such as 1D or 2D gel electrophoresis and, western blotting combined with colorimetric protein concentration determinations, functional characterization with activity assays using UV/VIS and various spectroscopies. For further characterization MALDI-TOF technique will be discussed in theory.
